Cyclen tetrahydrochloride is 1,4,7,10-tetraazacyclododecane with each of its four nitrogens protonated as the hydrochloride, formula C8H20N4·4HCl (C8H24Cl4N4), molecular weight 318.1 and CAS number 10045-25-7. It is a white to off-white solid. Free polyamines such as cyclen are strong bases that pick up water and carbon dioxide from the air, which makes the free base awkward to store, weigh and keep to specification. Converting it to the tetrahydrochloride gives a stable crystalline salt that can be purified by recrystallisation and dispensed accurately, and the free base is released with alkali at the point of use. The specification confirms both purity and stoichiometry: a chloride content of 40 to 45 percent corresponds to the full tetrahydrochloride, which is 44.6 percent chloride by theory, while the assay of 98.0 to 102.0 percent, single impurities at 0.05 percent and loss on drying at 5.0 percent control the organic purity. It is used as a stable source of cyclen for DOTA, DO3A and gadolinium contrast-agent synthesis, in preparing phosphinate, phosphonate and other macrocyclic chelators, and in coordination and radiopharmaceutical chemistry.
