DOTA, 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid, with the INN tetraxetan, has the formula C16H28N4O8, a molecular weight of 404.4 and CAS number 60239-18-1. It is a white to light yellow powder, made by attaching an acetic acid arm to each nitrogen of the cyclen ring. The four ring nitrogens and four carboxylate groups wrap around a metal ion to form a cage, and with gadolinium the result is one of the most thermodynamically and kinetically stable complexes known. That stability is the whole purpose: the gadolinium complex of DOTA, gadoteric acid, is formulated as its meglumine salt as a macrocyclic MRI contrast agent. DOTA itself is not a contrast agent; it is the ligand, and its purity determines how completely the metal is bound. It is also the standard anchor for radiometals such as gallium-68 and lutetium-177 in peptide-targeted radiopharmaceuticals. The material is usually a hydrate, so water, here up to 8.5 percent, should be taken into account when calculating the charge. It is used in the manufacture of gadoteric acid and gadoterate meglumine, in the synthesis of DOTA-based bifunctional chelators and radiopharmaceutical precursors, and in lanthanide coordination chemistry.
