Cobamamide, adenosylcobalamin or dibencozide, is a naturally occurring coenzyme form of vitamin B12, supplied as an orange-red crystalline powder. It carries a 5-deoxyadenosyl group at the sixth coordination position on the cobalt. The two active B12 forms serve different enzymes in different compartments of the cell, which is the basis for supplying both. Methylcobalamin works in the cytosol as the cofactor for methionine synthase in the methylation cycle. Cobamamide works in the mitochondria as the cofactor for methylmalonyl-CoA mutase, the enzyme that converts methylmalonyl-CoA to succinyl-CoA and thereby routes odd-chain fatty acids and several amino acids into the citric acid cycle for energy production. It is the predominant intracellular form of B12 in tissue, particularly in liver and muscle. A deficiency of this pathway is detected clinically by elevated methylmalonic acid, which is a more specific marker of B12 status than serum B12 itself. Like mecobalamin it is light-sensitive and requires protection from light throughout handling and in the finished pack. It is used in nutritional supplements, energy and sports formulations, pharmaceutical preparations, sublingual products and in veterinary nutrition.
